We ai'd our heifer on Friday and today we saw a spot of blood. Does this mean she didn't take or is it possible that she did conceive ?

There's often some blood a couple days after estrous. It has nothing to do with whether or not the cow settled. Take it as a sign that your heifer was in heat and your timing was good.
